Instructions for selecting pneumococcal antibody assays
Vaccine responses from pneumococcal vaccines are measured with the S-SpnAbVT and S-SpnAbNV antibody assays. The vaccines contain the following pneumococcal serotypes:
Pneumococcal polysaccharide vaccine:
- Pneumovax: 1, 2, 3, 4, 5, 6B, 7F, 8, 9N, 9V, 10A, 11A, 12F, 14, 15B, 17F, 18C, 19F, 19A, 20, 22F, 23F and 33F.
Pneumococcal conjugate vaccines:
- Synflorix: 1, 4, 5, 6B, 7F, 9V, 14, 18C, 19F and 23F
- Prevenar 13: 1, 3, 4, 5, 6A, 6B, 7F, 9V, 14, 18C, 19A, 19F and 23F
- Prevenar 7 (previously used): 4, 6B, 9V, 14, 18C, 19F and 23F
Measuring the vaccine response to a pneumococcal polysaccharide vaccine
The selection of the assay depends on whether the patient has received only a polysaccharide vaccine or also a conjugate vaccine in addition to the former (Figure 1).
The 6297 S-SpnAbVT assay is used to measure polysaccharide vaccine response in subjects who have not received a conjugate vaccine.
This antibody assay method measures response to the pneumococcal serotypes present in both polysaccharide and conjugate vaccines.
The 6296 S-SpnAbNV assay is used to measure a polysaccharide vaccine response in subjects who have received both a polysaccharide and a conjugate vaccine.
This antibody assay method measures response to the pneumococcal serotypes present only in polysaccharide vaccines.

Measuring vaccine response to pneumococcal conjugate vaccine
The 6297 S-SpnAbVT assay is used to measure vaccine response to pneumococcal conjugate vaccines (Figure 2).
The testing covers ten different pneumococcal serotypes, which are included in both the conjugate vaccine and the polysaccharide vaccine.
